If you are able to see an image of a slide under low objectives and such an image disappears under high power objectives, it means that the slide/image has not been centered properly.

What should be done would be to return the microscope to low power, center the image by adjusting the slide, and then go back to high power objective to view the image again.

When a slide containing a specimen is placed on the stage of a light microscope, such a slide should be adjusted so that the side containing the specimen is centered over the light coming from the condenser of the microscope.

Without centering before bringing the microscope into focus, the specimen's image will be skewed to one side of the field of vision just as we have in this case. Even though the microscope is parfocal, such an image will be out of focus at higher objectives.

Hence, the solution would be to ensure that images of specimens are centered at low power objectives before moving to high power.
